Illumina HiSeq and GAIIx

Illumina Sequencing

Illumina HiSeq
2000

We offer services on two Illumina Genome Sequencers (one HiSeq2000 and one GAIIx). GAIIx and HiSeq are short-read platforms. They can produce produce ~35 and ~150 million single reads per lane respectively. Paired-end and single read sequencing modes are available. For an overview of this technology please see Illumina Sequencing. We can produce the following read lengths for HiSeq: 50bp Single Read (SR), 50bp Paired End (PE), 75bp PE, 100bp SR, and 100bp PE. We can the following read lengths for GAIIx: 36bp SR, 75bp SR, 75bp PE, and 150bp PE. Barcoding is available for both instruments (free for Hiseq and additional cost for GAIIx). Because of decrasing demand on our GAIIx and the impossiblity to find partners to complete flowcells, we only accept full flowcell sequencing orders for GAIIx. However, you can submit one lane at the time for the HiSeq. Note that some HiSeq sequencing modes are more popular than others (i.e. easier to find partners to complete flowcells). At the moment the two most popular sequencing modes are: 50bp SR, 100bp PE, and to a lesser degree 50bp PE. 100bp SR and 75bp PE are rarely requested. When you submit samples for sequencing, the waiting time will be a combination of the number of libraries you've requested, the time it will take to find partners if you've requested a partial flowcell (7 lanes for a full flowcell), and the instrument queue (see instrument queues).

Sample Requirements

Samples must be resuspended in TE or nuclease-free water. Samples should be submitted in an eppendorf tube clearly marked with a 2- to 3- digit identifier. See input amount in the table below.

Library Type Input Concentration

Volume

DNA-Seq DNA 40 ng/µl 50 µl
RNA-Seq total RNA 20 ng/µl 50 µl
ChIP-Seq ChIP enriched DNA 1 ng/ul 30 µl
smRNA-Seq total RNA 20 ng/µl 50 µl
Mate-Pair DNA 200 ng/µl 50 µl

Note that Nanodrop frequently over-estimate DNA concentration (~3x).
