We offer services on two Roche 454 GS-FLX+ sequencinger. 454 Sequencing is a long-read platform. This technology is often used for these applications: De novo genome or transcriptome sequencing, amplicon sequencing, genome or transcriptome resequencing, and metagenomics. It can produce more than a million reads at an >550bp average length for genome and transcriptome sequencing (FLX+ chemistry) and >375bp average length for amplicon and paired end sequencing (Titanium chemistry).
Samples must be resuspended in TE or nuclease-free water. Samples should be submitted in an eppendorf tube clearly marked with a 2- to 3- digit identifier. See input amount in the table below.
| Library type | Input | Quantity |
Volume |
|---|---|---|---|
| gDNA | DNA | 2 µg | 110 µl |
| Amplicon | DNA | 50 µl | |
| cDNA | total RNA, RIN>7 | 50-75 µg | 50 µl |
| Paired-end (3kb, 8kb, 20kb) | DNA | 5, 15, 30 µg | 110 µl |
